Vitamin B6 P-5-P and B12 methylcobalamin: active forms and their real role

In brief. Pyridoxal-5-phosphate (P-5-P/PLP) is a coenzyme form of vitamin B6; methylcobalamin is a biologically active form of vitamin B12. These terms describe specific molecules, but they do not prove universal clinical superiority. The NIH states that there is no evidence that vitamin B12 absorption from supplements varies by form. What is P-5-P?

Vitamin B6 exists in several forms. Pyridoxal-5-phosphate is a coenzyme form involved in many enzymatic reactions. Naming P-5-P specifies the form present in the formula; by itself, it does not demonstrate better clinical outcomes than another B6 source.

What is methylcobalamin?

Vitamin B12 includes several cobalamins. Methylcobalamin and adenosylcobalamin are coenzyme forms used by the body. Cyanocobalamin and hydroxocobalamin can be converted into active forms.

The NIH Office of Dietary Supplements states that no evidence shows that absorption rates of supplemental B12 vary by form. The available data therefore do not support saying that methylcobalamin is globally superior to other forms for every user.

What can be said about vitamins B6 and B12 under EU regulation?

When content conditions are met, vitamins B6 and B12 have authorized European claims including contribution to normal energy-yielding metabolism, normal functioning of the nervous system, normal psychological function and reduction of tiredness and fatigue.

These claims apply to the nutrient, not to the superiority of P-5-P or methylcobalamin, and not to the clinical efficacy of Régénère™ or Ressource™.

“Methylated vitamins”: what does the term really mean?

Marketing often groups 5-MTHF and methylcobalamin under this phrase. Methylation is a real set of biochemical reactions, but generic symptoms cannot diagnose a “methylation problem”. A methylated form is not, by itself, a treatment for a genetic polymorphism or a personalized recommendation.

Why document the form if it is not necessarily “better”?

Because raw-material identity is useful information. Transparency does not mean automatic hierarchy. Naming the form tells you what is actually in the capsule, then allows that fact to be separated from efficacy claims, which require their own evidence.
